Pharmacological action
Synthetic analog of histamine. It is a weak agonist of histamineH1-receptors and a fairly powerful antagonist of histamineH3-receptors. It has a histamine-like and vasodilating effect. It affects the cochlear blood flow and the central vestibular apparatus.
The effect of the drug includes vasodilation in the inner ear (mediated through histamineH3 and H1receptors), an inhibitory effect on the vestibular nuclei (through histamineH3receptors) and the pulse activity of ampullary receptors. By direct agonistic action on histamineH1-receptors of the inner ear vessels and precapillary sphincters of the microcirculatory bed located in the vascular strip (stria vascularis), as well as indirectly through histamineH3-receptors, it improves microcirculation and capillary permeability of the inner ear, normalizes endolymph pressure in the labyrinth and cochlea, increases blood flow in the basilar artery.
It has a pronounced central effect, being an antagonist of histamineH3-receptors of the vestibular nerve nuclei. Normalizes the conduction in polysynaptic neurons of the vestibular nuclei at the level of the brain stem. Indirectly acting on histamineH3receptors, it increases the content of serotonin in the brain stem, which reduces the activity of the vestibular nuclei.
Helps to eliminate disorders of both the vestibular and cochlear apparatus: reduces the frequency and intensity of dizziness, reduces tinnitus, improves hearing in cases of its decline.
Betahistine stimulates histamineH1receptors, so it does not have a sedative effect and does not cause drowsiness.
