Composition
5 ml of syrup contains:
Loratadine 5 mg
. excipients:
Citric acid,
artificial flavor (peach),
glycerol,
propylene glycol,
sodium benzoate,
granulated sucrose,
water (pH 25-31).
Pharmacological action
Claritine is an antihistamine, a blocker of peripheral histamine H1-receptors. It is characterized by selective antagonism towards peripheral receptors of this type. When conducting skin tests for histamine after taking one (10 mg) or several doses of Claritine, the antihistamine effect was manifested in 1-3 hours, reached a maximum in the period from 8 to 12 hours from the onset of action, and lasted more than 24 hours.
When taking Claritine for 28 days, there was no development of resistance to the drug.
In a clinical study in which Claritine was administered for 90 days at a dose 4 times higher than the therapeutic dose, no clinically significant prolongation of the QT interval was detected on the ECG.
Indications
– treatment of seasonal and year-round allergic rhinitis and elimination of symptoms associated with these diseases: sneezing, itching of the nasal mucosa, rhinorrhea, burning sensation and itching in the eyes –
– treatment of skin diseases of allergic origin (including chronic urticaria) in adults and children aged 2 years.
Use during pregnancy and lactation
The use of the drug during pregnancy is possible only in cases of extreme necessity.
No teratogenic effects of loratadine were found in experimental animal studies.
Loratadine and its metabolite descarboethoxyloratadine easily pass into breast milk, where they reach concentrations close to their plasma concentrations.
Therefore, if it is necessary to use the drug during lactation, you should stop breastfeeding.
Contraindications
-lactation (breast-feeding); – hypersensitivity to loratadine or other components of Claritine.
The drug should be used with caution during pregnancy.
Side effects
From the digestive system in adults: possible-dry mouth, nausea, gastritis; rarely-liver function disorders. From the side of the central nervous system in adults: possibly-headache, fatigue, drowsiness; in children (rarely) – headache, nervousness, sedation.
Allergic reactions in adults: possible skin rash; rarely-anaphylactic reactions.
Dermatological reactions in adults: there have been rare reports of alopecia.
The frequency of these side effects with Claritine was at the same level as with placebo.
Interaction
When Claritine was co-administered with ketoconazole, erythromycin, or cimetidine, an increase in the concentration of loratadine and its metabolite in blood plasma was noted, which was not clinically manifested (including according to ECG data).
How to take, course of use and dosage
For adults, elderly patients and children over 12 years of age, the recommended dose of Claritine is 2 teaspoons (10 ml) of syrup 1 time/day.
For children aged 2 to 12 years, the recommended dose of Claritine is 5 mg – 1 teaspoon (5 ml) of syrup if the body weight is less than 30 kg. ; with a body weight of 30 kg or more-10 mg-2 teaspoons (10 ml) of syrup 1 time/day.
For patients with impaired liver function or renal insufficiency (creatinine clearance less than 30 ml / min) the initial dose of the drug is 10 mg – 2 teaspoons (10 ml) of syrup every other day.
Overdose
Symptoms: in adult patients, headache, drowsiness and tachycardia were observed when taking the drug in doses of 40-180 mg, significantly exceeding the recommended therapeutic dose of 10 mg. Extrapyramidal symptoms and palpitations were observed in children with a body weight of less than 30 kg when taking Claritine syrup at a dose of more than 10 mg.
Treatment: conducting symptomatic and supportive care. Loratadine is not eliminated from the body during hemodialysis. There are no data on the elimination of loratadine during peritoneal dialysis.

Special instructions
Claritine should be discontinued no later than 48 hours prior to skin testing to avoid distortion of the results. In the study of psychomotor function after the use of Claritine simultaneously with alcohol, the potentiating effect of Claritine on alcohol was not revealed.
Use in pediatrics the drug should be used in children under 2 years of age.
