Pharmacological action

Sedative.

A combined drug whose action is determined by the properties of its constituent substances. It has a sedative and antispasmodic effect, moderate cardiotonic effect (slows down the rhythm and increases the strength of heart contractions), has moderate hypotensive properties.

Ethylbromizovalerianate has a sedative and antispasmodic effect due to irritation, mainly of the oral and nasopharyngeal receptors, a decrease in reflex excitability in the central nervous system and increased inhibition in cortical neurons and subcortical structures of the brain, as well as a decrease in the activity of central vasomotor centers and a direct local antispasmodic effect on smooth muscles. By the nature of its action, it is close to valerian preparations.

Motherwort herb has a pronounced sedative effect (calming), moderately reduces the frequency and increases the strength of heart contractions, shows moderate hypotensive properties.

Peppermint oil has a reflex vasodilating, antispasmodic, mild choleretic, antiseptic effect. The mechanism of action is associated with the ability to irritate the “cold” receptors of the oral mucosa and reflexively expand mainly the vessels of the heart and brain. Eliminates the phenomena of flatulence due to irritation of the mucous membrane receptors of the gastrointestinal tract (GIT), increasing intestinal motility.

Diphenhydramine is a first-generation H1-histamine receptor blocker. The effect on the central system is caused by the blockade of H1-histamine receptors of m-holinoreceptors of the brain. Reduces or prevents histamine-induced smooth muscle spasms, increased capillary permeability, tissue edema, itching and hyperemia, has antiemetic, sedative effects, and has a hypnotic effect. Antagonism with histamine is manifested to a greater extent in relation to local vascular reactions than to systemic ones, that is, a decrease in blood pressure.

Indications

• Increased nervous excitability;
• as a sedative for sleep disorders that are not associated with organic disorders of the central nervous system;
• neurasthenia.

Use during pregnancy and lactation

Pregnancydifenhydramine is able to pass through the placenta; cases of jaundice and extrapyramidal disorders have been described in children born to women who took diphenhydramine during pregnancy. Taking diphenhydramine in the third trimester can lead to adverse reactions in the newborn or premature birth. The safety of Corvalol Neo during pregnancy has not been established and its use during this period is contraindicated. Breast-feedingapplication of diphenhydramine during breastfeeding can cause increased agitation and irritability of the child. If you need to use Corvalol Neo during breast-feeding, you should decide whether to stop breastfeeding.

Contraindications

– stenosis of the bladder neck;

– bronchial asthma;

– epilepsy;

– pregnancy;

– lactation (breastfeeding);

– children’s age up to 18 years (efficacy and safety not established);

– porphyria;

– pyloroduodenal stenosis;

erosive-ulcerative lesions of the gastrointestinal tract (in the phase of exacerbation);

– stenosing peptic ulcer of stomach and duodenum.

With prolonged use, resistance may develop. The drug may increase the effect of alcohol, so alcohol intake during treatment should be excluded.

Use in combination with other antihistamines, including topical medications, cough and cold remedies, should be avoided.

Side effects

Side effects are more likely to develop in elderly patients. Use in elderly patients with confused consciousness should be avoided.

Diphenhydramine may worsen existing tinnitus.

Corvalol Neo contains ethyl alcohol in its composition (see the section “Special instructions”), so the drug should be used with caution in alcoholism, as well as in high-risk patients, including those with liver diseases, traumatic brain injuries, and brain diseases.

If you have one of these diseases/If you have any serious medical conditions, be sure to consult your doctor before taking the drug.

Blood and lymphatic system disorders: hemolytic anemia, thrombocytopenia, agranulocytosis.

Immune system disorders: allergic reactions, including rash, hives, difficulty breathing, angioedema.

Skin and subcutaneous tissue disorders: photosensitivity.

Mental disorders: confusion, paradoxical agitation (especially affected by elderly patients); in some cases, irritability, euphoria.

Nervous system disorders: drowsiness, weakness, decreased speed of psychomotor reactions, impaired coordination and reaction speed, fatigue, dizziness, decreased ability to concentrate, instability, convulsions, headache, paresthesia, dyskinesia; in some cases, insomnia, tremor.

Visual disturbances: blurred vision.

Hearing and labyrinth disorders: tinnitus.

Disorders of the cardiovascular system: low blood pressure, slow heart rate, tachycardia, extrasystole.

Disorders of the respiratory system, chest and mediastinal organs: dryness of the nasal mucosa, bronchi (thickening of bronchial secretions).

From the gastrointestinal tract: dry mouth, dyspepsia, nausea, vomiting, epigastric discomfort, anorexia.

Muscle, skeletal and connective tissue disorders: muscle twitching.

Disorders of the kidneys and urinary tract: difficulty urinating, urinary retention.

These symptoms disappear with a decrease in the dose of the drug or discontinuation of the drug. With prolonged use of high doses of the drug, drug dependence may develop, as well as bromine accumulation in the body and the development of bromism, the manifestations of which are: depressive mood, apathy, rhinitis, conjunctivitis, hemorrhagic diathesis, impaired coordination of movements.

Interaction

When the drug Corvalol Neo is used simultaneously with ethanol and drugs that inhibit the activity of the central nervous system (sedatives, sleeping pills, antipsychotics, tranquilizers, tricyclic antidepressants), mutual strengthening of the effects is possible.

Monoamine oxidase (MAO) inhibitors increase the severity and duration of the anticholinergic effects of diphenhydramine. Corvalol Neo should be taken together with MAO inhibitors with caution or 2 weeks after their withdrawal.

Diphenhydramine may increase the anticholinergic effects of other medications (atropine, tricyclic antidepressants).

Antagonistic interaction is noted when co-administered with psychostimulants.

Diphenhydramine is an inhibitor of the cytochrome P450 isoenzyme CYP2D6. Thus, Corvalol Neo can potentially interact with drugs whose primary metabolism is carried out by CYP2D6.

Corvalol Neo enhances the effect of antispasmodic, analgesic agents.

Alcohol increases the effect of the drug and its toxicity.

Reduces the effectiveness of apomorphine as an emetic in the treatment of poisoning.

How to take it, course of administration and dosage

Take orally, before meals,3 times a day for 30 drops, previously dissolved in a small amount (30-40 ml) of water. Shake the bottle before use.

The course of treatment is 10-15 days. On the recommendation of a doctor, a second course of treatment can be carried out, but not earlier than in 1 month.

Overdose

Symptoms:

Central nervous system depression, agitation or depression, dilated pupils, dry mouth, gastrointestinal paresis, fever, hot flashes, tremors, dystonic reactions, hallucinations, and ECG changes. A significant overdose can cause rhabdomyolysis, seizures, delirium, toxic psychosis, arrhythmias, coma, and cardiovascular failure.

Treatment for overdose:

There is no special antidote.

Gastric lavage.If necessary – symptomatic treatment: drugs that increase blood pressure, oxygen, intravenous use of plasma-substituting fluids.

Do not use epinephrine or analeptics.

If you experience symptoms of overdose, you should stop taking Corvalol Neo and immediately consult a doctor.

Transparent liquid of greenish-brown color with a characteristic smell.

During storage, turbidity and sediment formation in the form of particles and/or flakes are allowed, during shaking of which a suspension of small particles is formed.

Special instructions

There is no experience of using the drug in children under 18 years of age, during pregnancy and during breastfeeding.

During the use of the drug, avoid UV radiation and ethanol intake.

Long-term use of the drug in high doses is not recommended, due to the possible development of overdose.

Due to the possible toxic effect of bromine accumulation in the body, long-term use in high doses is not recommended.

It is necessary to inform the doctor about the use of this drug: the antiemetic effect may make it difficult to diagnose appendicitis and recognize symptoms of overdose with other medications.

Corvalol Neo contains 47.5% ethyl alcohol, so when taken orally in the maximum single dose of the drug, the content of absolute ethyl alcohol (ethanol) is 0.308 g, in the maximum daily dose – 0.923 g.

Influence on the ability to drive vehicles and mechanisms:

Due to the possibility of dizziness, decreased concentration of attention and other side effects associated with taking the drug, during treatment, patients are advised to refrain from driving vehicles and engaging in other potentially dangerous activities that require increased concentration of attention, speed of psychomotor and motor reactions.
