Pharmacodynamics : Guaifenesin has a stimulating effect on the receptors of the gastric mucosa, leading to increased secretion of the glands of the gastrointestinal tract (GIT) and a reflex increase in the secretion of the glands of the respiratory tract. As a result, the viscosity decreases and the volume of bronchial secretions increases. Other effects may include stimulation of the vagus nerve endings in the bronchial glands and stimulation of certain brain centers, which in turn leads to improved sputum discharge. Guaifenesin has an expectorant effect for 24 hours. The components that make up the drug have a warming, softening and soothing effect when swallowed. Pharmacokinetics: Guaifenesin is well absorbed from the gastrointestinal tract when taken orally. In healthy adult volunteers, the maximum blood concentration (Cmax) and time to reach maximum blood concentration (TMAX) after ingestion of 600 mg of guaifenesin were approximately 1.4 mcg/ml and 15 minutes, respectively. The half-life (T 1/2) was about 1 hour, the drug was not detected in the blood after about 8 hours. Guaifenesin is metabolized in the liver. It is mainly excreted by the kidneys.
