Pharmacological action

H1-histamine receptor blocker (long-acting).

Inhibits the release of histamine and leukotriene C4 from mast cells. Prevents the development and eases the course of allergic reactions.

It has anti-allergic, antipruritic, and antiexudative effects. Reduces capillary permeability, prevents the development of tissue edema, and relieves smooth muscle spasms.

The anti-allergic effect develops in 30 minutes, reaches a maximum in 8-12 hours and lasts for 24 hours. It does not affect the central nervous system (because it does not penetrate the BBB) and is not addictive.

Indications

Allergic rhinitis (seasonal and year-round), pollinosis, allergic conjunctivitis, chronic idiopathic urticaria, angioedema, itchy dermatoses, pseudoallergic reactions caused by histamine release, allergic reactions to insect bites.

Contraindications

Hypersensitivity, lactation period.

Side effects

Nervous system disorders:  anxiety, agitation (in children), asthenia, drowsiness, hyperkinesia, paresthesia, tremor, amnesia, depression, increased fatigue.

Skin disorders: dermatitis.

From the genitourinary system: discoloration of urine, painful urge to urinate-emission; dysmenorrhea, menorrhagia, vaginitis.

From the side of metabolism:  weight gain, sweating, thirst.

From the musculoskeletal system:  cramps of the calf muscles, arthralgia, myalgia.

From the digestive system:  dry mouth, taste changes, anorexia, constipation or diarrhea, dyspepsia, gastritis, flatulence, increased appetite, stomatitis, nausea, vomiting.

Respiratory system disorders:  cough, bronchospasm, dryness of the nasal mucosa, sinusitis.

From the side of the senses:  visual impairment, conjunctivitis, pain in the eyes and ears.

Interaction

CYP3A4 inhibitors (including ketoconazole, erythromycin), CYP3A4 and CYP2D6 inhibitors (cimetidine, etc. ) increase the concentration of loratadine in the blood.

How to take, course of use and dosage

Adults and children with a body weight of more than 30 kg – 10 mg 1 time a day.

In case of hepatic insufficiency, the initial dose is 5 mg / day.

Children from 2 to 12 years of age with a body weight of less than 30 kg – 5 mg / day in 1 reception.

Overdose

Drowsiness, tachycardia, headache.

In case of overdose, you should consult a doctor and take measures to remove the drug from the gastrointestinal tract and reduce absorption (emetics, gastric lavage, laxatives, activated charcoal).

Peritoneal dialysis is ineffective, and hemodialysis is not recommended.

Special instructions

During the treatment period, care should be taken when driving vehicles and doing other activities. potentially dangerous activities that require increased concentration of attention and speed of psychomotor reactions due to the risk of drowsiness.

 In the future, the degree of restriction is set individually for each patient.
