of Phencarol – antiexudative, antipruritic, antiallergic, decongestant.
Pharmacodynamics
The Active ingredient of the drug hifenadine-a blocker of H1-histamine receptors-prevents the development and facilitates the course of allergic reactions. It has an anti-allergic, anti-exudative and antipruritic effect, preventing the development of allergic inflammation in the tissue.
Weakens the effect of histamine, reduces its effect on vascular permeability (reducing permeability, has an anti-edematous effect), reduces its bronchospastic effect and spasmogenic effect on intestinal smooth muscle, weakens the hypotensive effect of histamine.
Hifenadine reduces the content of histamine in tissues (due to the ability to activate diamine oxidase, an enzyme that inactivates histamine). During the course of treatment, the antihistamine effect of hifenadine is not reduced. It has a moderate antiserotonin effect, shows weak m-holinoblokiruyuschuyu activity.
Pharmacokinetics
45% of hifenadine is rapidly absorbed from the gastrointestinal tract and is detected in body tissues within 30 minutes. Cmax of Active ingredient in blood plasma is reached after 1 h. It has low lipophilicity, poorly penetrates the BBB.
The highest content of the Active ingredient was observed in the liver, slightly lower in the lungs and kidneys, and the lowest in the brain (less than 0.05%, which explains the absence of a depressing effect on the central nervous system). Hifenadine is metabolized in the liver. Metabolites are excreted by the kidneys and intestines. The unabsorbed part of the drug is removed from the intestine.
Indications
- Various manifestations of allergies;
- neurodermatitis;
- pollinosis;
- acute and chronic urticaria;
- angioedema;
- allergic rhinitis;
- dermatoses, including eczema, psoriasis, neurodermatitis, pruritus.
Contraindications
- peptic ulcer of the stomach and duodenum in the acute phase

- disorders hypersensitivity to Phencarol.
Side effects
From the digestive system: dryness of the oral mucosa, nausea, vomiting.
From the central nervous system: drowsiness, headache.
Other: allergic reactions.
Interaction
It does not enhance the depressing effect of alcohol and sleeping pills on the central nervous system.
Having weak M-cholinoblocking properties, it can reduce the motility of the gastrointestinal tract and increase the absorption of slowly absorbed medications (for example, indirect anticoagulants-coumarins).
How to take, course of use and dosage
The drug should be taken orally, after meals. The course of treatment for adults is 10-20 days, if necessary – again. The duration of treatment for children is 10-15 days.
Recommended dosages: adults-25-50 mg 3-4 times a day (the maximum daily dose is 200 mg); children from 2 to 3 years-5 mg 2-3 times a day; children 3-7 years-10 mg 2 times a day; children 7-12 years-10-15 mg 2-3 times a day.
Overdose
Symptoms: dryness of the mucous membranes, headache, vomiting, abdominal pain and other dyspeptic phenomena.
Treatment: symptomatic.
It is necessary to flush the stomach, take activated charcoal, and immediately consult a doctor.
Special instructions
The absence of a pronounced anticholinergic blocking effect allows prescribing antihistamines with anticholinergic activity to patients who are contraindicated.
Influence on the ability to drive motor vehicles and manage mechanisms: persons whose profession requires rapid mental and motor reactions should first determine (by short-term use) whether the drug has a sedative effect.
