Composition

Tablets 1 tab. loratadine 10 mg

Pharmacological action

Histamine H1-receptor blocker.

It has anti-allergic, antipruritic, and antiexudative effects.

Reduces capillary permeability, prevents the development of tissue edema, reduces the increased contractile activity of smooth muscles due to the action of histamine.

Indications

Seasonal and year-round allergic rhinitis, conjunctivitis, acute urticaria and angioedema, histaminergic symptoms caused by the use of histaminoliberates (pseudoallergic syndromes), allergic reactions to insect bites, complex treatment of itchy dermatoses (contact allergodermatitis, chronic eczema).

Contraindications

Pregnancy, lactation, children under 2 years of age, hypersensitivity to loratadine.

Side effects

From the digestive system: rarely – dry mouth, nausea, vomiting, gastritis; in some cases-liver function disorders.

From the central nervous system: rarely – increased fatigue, headache, excitability (in children).

From the cardiovascular system: rarely-tachycardia.

Allergic reactions: rarely-skin rash; in isolated cases-anaphylactic reactions.

Dermatological reactions: in some cases – alopecia.

Interaction

When loratadine is co-administered with drugs that inhibit the CYP3A4 and CYP2D6 isoenzymes or are metabolized in the liver with their participation (including cimetidine, erythromycin, ketoconazole, quinidine, fluconazole, fluoxetine), it is possible to change the plasma concentration of loratadine and/or these drugs.

Inducers of microsomal oxidation (phenytoin, ethanol, barbiturates, zixorin, rifampicin, phenylbutazone, tricyclic antidepressants) reduce the effectiveness.

How to take, course of use and dosage

Inside adults and children over 12 years of age, as well as with a body weight of more than 30 kg-10 mg 1 time/day.

Children from 2 to 12 years of age with a body weight of less than 30 kg – 5 mg 1 time/day.

Overdose

Symptoms: headache, drowsiness, tachycardia. Extrapyramidal disorders and palpitations were observed in children weighing less than 30 kg when ingested at a dose of more than 10 mg.

Treatment: gastric lavage, taking activated charcoal; symptomatic and supportive therapy. For children,0.9% sodium chloride solution is used for washing, for adults, water can be used for washing. Saline laxative solutions and enterosorbents are indicated for rapid elimination of loratadine from the gastrointestinal tract. Loratadine is not eliminated by hemodialysis or peritoneal dialysis.

Special instructions

When using loratadine, the development of seizures cannot be completely excluded, especially in predisposed patients.

Patients with impaired renal or hepatic function need to adjust the dosage regimen.
