Pharmacological action
Magnerot is a magnesium preparation. Magnesium is a macronutrient, essential for many energy processes, and is involved in the metabolism of proteins, fats, carbohydrates, and nucleic acids.
Magnesium takes part in the process of neuromuscular excitation, inhibiting neuromuscular transmission. Magnesium is of particular interest as a natural physiological calcium antagonist. Magnesium controls the normal functioning of myocardial cells, participates in the regulation of contractile function of the myocardium. In stressful situations, an increased amount of free ionized magnesium is removed, and therefore an additional amount of magnesium contributes to an increase in stress resistance.
Magnesium deficiency leads to neuromuscular disorders (motor and sensory hyperexcitability, seizures, paresthesia), mental changes (depression, confusion and hallucinations), cardiovascular diseases (ventricular extrasystole, tachycardia, hypersensitivity to cardiac glycosides). Magnesium deficiency during pregnancy increases the likelihood of toxicosis and premature birth. Orotic acid salts are involved in the process of metabolism. In addition, orotic acid salts are necessary for the fixation of magnesium on ATP in the cell and the manifestation of its action.
Pharmacokinetics
Approximately 35-40% of the dose is absorbed. Hypomagnesemia stimulates the absorption of magnesium ions. The presence of orotic acid salts improves the absorption of magnesium. Magnesium is excreted by the kidneys, and excretion decreases when magnesium is deficient and increases when it is excess.
Indications
Established magnesium deficiency, isolated or associated with other deficient conditions, accompanied by symptoms such as increased irritability, minor sleep disturbances, gastrointestinal spasms, increased fatigue, muscle aches and spasms, and tingling sensation in the muscles. If after a month of treatment there is no reduction in these symptoms, continuation of treatment is impractical.
Use during pregnancy and lactation
It is possible to use the drug during pregnancy and lactation (breastfeeding )according to indications, since during these periods the need for magnesium increases significantly.
If its content is not balanced, it can lead to serious complications, including miscarriage.
The use of the drug during pregnancy and lactation is possible if the potential benefit to the mother exceeds the potential risk to the fetus and child.
Contraindications

Interaction
Concomitant use of magnesium-containing drugs with iron, sodium fluoride and tetracyclines may decrease the absorption of the latter. In addition, aminoquinolines, quinidine, penicillamine should not be administered simultaneously with magnesium preparations. Therefore, you should take these medications at intervals of 2-3 hours. Oral contraceptives, diuretics, muscle relaxants, glucocorticosteroids, and insulin reduce the effect of the drug. Magnesium should be used with caution if you are taking magnesium-containing medications, such as antacids or laxatives, or potassium-sparing diuretics or medications containing calcium. When used concomitantly with drugs containing aluminum, the absorption of aluminum in the body can be increased. Magnesium should be used with caution when used concomitantly with digitalis preparations, as concomitant use may impair the absorption of digitalis preparations. Some antibiotics (aminoglycosides), cisplatins, and cyclosporine A may lead to increased magnesium excretion. Some diuretics (thiazides and furosemide), cetuximab and erlotinib, proton pump inhibitors (omeprazole and pantoprazole), foscarnet, pentamidine, rapamycin and amphotericin B may cause magnesium deficiency and thus reduce the effect of Magnerot®.
How to take, course of use and dosage
Inside, before meals, with a small amount of liquid.
Assign 2 tablets 3 times a day for 7 days, then-1 tablet 2-3 times a day daily.
The course duration is at least 4-6 weeks. If necessary, Magnerot® can be used for a long time.
For nocturnal cramps of the calf muscles, it is recommended to take 2-3 tablets in the evening.
Overdose
With normal renal function, ingestion of magnesium does not cause toxic reactions. Magnesium poisoning can occur with kidney failure. Toxic effects mainly depend on the concentration of magnesium in the blood serum. Symptoms: low blood pressure, nausea, vomiting, depression, slow reflexes, respiratory depression, coma, cardiac arrest, anuric syndrome. Treatment: rehydration, forced diuresis, with renal failure, hemodialysis or peritoneal dialysis is necessary.
