Composition

Active ingredient:

clemastine 1 mg, which is equivalent to 1.34 mg of clemastine hydrofumarate.

Auxiliary substances:

lactose monohydrate,

povidone (polyvinylpyrrolidone),

corn starch,

magnesium stearate, talc.

Pharmacological action

Histamine H1-receptor blocker, ethanolamine derivative. It has a strong antihistamine and antipruritic effect with a rapid onset of action and a duration of up to 12 hours, prevents the development of vasodilation and contraction of smooth muscles induced by histamine. Having an anti-allergic effect, it reduces the permeability of blood vessels, capillaries, inhibits exudation and edema formation, reduces itching, and has a m-holinoblocking effect.

Clinical pharmacology

Absorption After oral use, clemastine is almost completely absorbed from the gastrointestinal tract. Cmax in blood plasma is reached in 2-4 hours. Distribution The binding of clemastine to plasma proteins is 95%. It is excreted in breast milk in small amounts. Metabolism and elimination from plasma is biphasic, the corresponding T1/2 is 3.6 ± 0.9 h and 37 ± 16 h. Clemastine undergoes significant metabolism in the liver. Metabolites are mainly (45-65%) excreted in the urine; unchanged Active ingredient is found in the urine only in trace amounts.

Indications

• Pollinosis (hay fever, including allergic rhinoconjunctivitis);
• urticaria of various origins;
• pruritus, itchy dermatoses;
• acute and chronic eczema, contact dermatitis;
• drug allergy;
• insect bites.

Use during pregnancy and lactation

The use of the drug during pregnancy and lactation is contraindicated.

Contraindications

• Diseases of the lower respiratory tract (including bronchial asthma);
• simultaneous use of MAO inhibitors;
• children under 6 years of age;
• pregnancy;
• lactation (breastfeeding);
• hypersensitivity to the components of the drug.

Tavegil should be used with caution in patients with stenosing gastric ulcer, pyloroduodenal obstruction, bladder neck obstruction, as well as prostatic hypertrophy accompanied by urinary retention, increased intraocular pressure, hyperthyroidism, and diseases of the cardiovascular system (including arterial hypertension).

Side effects

Determination of the frequency of adverse reactions: very common (≥1/10); common (≥1/100, <1/10); infrequent (≥1/1000, <1/100); rare (≥1/10 000, <1/1000); very rare (From the nervous system: often-increased fatigue, drowsiness, sedation, weakness, fatigue, lethargy, impaired coordination of movements; infrequently-dizziness; rarely-headache, tremor, stimulating effect. From the digestive system: rarely-dyspepsia, nausea, vomiting, gastralgia; very rarely-constipation, dry mouth; in some cases – decreased appetite, diarrhea. From the respiratory system: rarely-thickening of bronchial secretions and difficulty separating sputum, a feeling of pressure in the chest and difficulty breathing, nasal congestion. From the cardiovascular system: rarely-low blood pressure (more often in elderly patients), extrasystole; very rarely-palpitations. From the side of the senses: rarely-impaired clarity of visual perception, diplopia, acute labyrinthitis, tinnitus. From the urinary system: very rarely-frequent urination, difficulty urinating. From the hematopoietic system: rarely-hemolytic anemia, thrombocytopenia, agranulocytosis. Dermatological reactions: rarely – skin rash, photosensitization. Allergic reactions: rarely-anaphylactic shock.

Interaction

Tavegil ® potentiates the action of drugs that depress the central nervous system (sleeping pills, sedatives, tranquilizers), m-holinoblokatorov, as well as ethanol. Incompatible with concomitant use of MAO inhibitors.

How to take, course of use and dosage

Inside, adults and children over 12 years of age are prescribed 1 tab. (1 mg) morning and evening. In cases that are difficult to treat, the daily dose can be up to 6 tablets. (6 mg). Children aged 6-12 years are prescribed 1/2-1 tab. before breakfast and at night. Tablets should be taken before meals, washed down with water.

Overdose

Symptoms: overdose of antihistamines can lead to both depressing and stimulating effects on the central nervous system. CNS stimulation is more common in children. Manifestations of anticholinergic action may also develop: dry mouth, fixed dilation of the pupils, flushes of blood to the upper half of the trunk, gastrointestinal disorders (nausea, epigastric pain, vomiting). Treatment: if the patient does not vomit spontaneously, then it should be induced artificially (only if the patient’s consciousness is preserved). If 3 hours or a little more have passed since taking the drug, gastric lavage can be performed using an isotonic sodium chloride solution. You can also prescribe a saline laxative. Symptomatic therapy is also indicated.

Special instructions

To prevent distortion of the results of skin scarification tests for allergens, the drug should be discontinued 72 hours before the allergological testing. The tablets contain lactose, so the drug is not recommended for patients suffering from rare congenital diseases associated with galactose intolerance, severe lactase deficiency and glucose-galactose malabsorption. Use in pediatrics Avegil® in tablet form is contraindicated in children under 6 years of age. For the treatment of children aged from 1 year, Tavegil® can be used in the form of a solution for intravenous and intramuscular use. Effects on the ability to drive motor vehicles and manage Mechanicsmiclemastine has a slight sedative effect (from weak to moderate in intensity), so patients taking Tavegil® are recommended to refrain from driving vehicles, working with mechanisms, as well as from other activities that require increased attention and speed of psychomotor reactions.
