Pharmacological action
Thiotriazoline is a drug with pronounced membrane-stabilizing, antioxidant, reparative and immunomodulatory activity. Thiotriazoline also has some anti-ischemic effects.
The mechanism of action of thiotriazoline is associated with the ability of the drug to enhance compensatory activation of anaerobic glycolysis and activate the processes of oxidation in the Krebs cycle without changing the intracellular ATP Fund.
Because of a molecule of tertiary thiol sulfur and nitrogen thiotriazolin activates the antioxidant system, which reduces the negative impact of free radicals, as well as the intensity of peroxidation and formation of free oxygen ions.
Thiotriazoline reduces the sensitivity of the myocardium to catecholamines, prevents progressive inhibition of the contractile function of the myocardium, and also helps to reduce the area of ischemia and myocardial necrosis.
In addition, thiotriazoline normalizes metabolic processes in the myocardium, contributes to the normalization of heart rhythm and improves the rheological properties of the blood by activating the fibrinolytic system.
Thiotriazoline has a pronounced hepatoprotective activity, prevents the destruction of hepatocytes and stimulates their regeneration, helps to reduce the spread of necrosis and fatty liver infiltration.
Thiotriazoline normalizes protein, pigment, carbohydrate and lipid metabolism in hepatocytes. It has some choleretic effect and normalizes the composition of bile.
When administered orally, thiotriazoline is well absorbed, with a bioavailability of up to 53%. With a single oral use, the peak plasma concentration of thiotriazoline is reached within 1.6 hours, with intramuscular use-0.84 hours, and intravenous use-0.1 hours.
The elimination half-life is about 8 hours. Thiotriazoline accumulates in the tissues of the kidneys and to a small extent in the tissues of the lungs, colon, heart and spleen.
Indications
- Chronic alcoholic hepatitis of minimal and moderate activity.
- Chronic viral hepatitis, fatty liver disease, cirrhosis of the liver.
- Toxic liver diseases, including those of alcoholic, viral, and medicinal origin (antibiotics, antitumor, antituberculous, antiviral drugs, tricyclic antidepressants, oral contraceptives, etc. ).
Use during pregnancy and lactation
Contraindicated during pregnancy and lactation.

Side effects
Thiotriazoline is generally well tolerated. Isolated cases of adverse reactions have been observed with the clinical use of Thiotriazoline:
From the gastrointestinal tract: nausea, dry mouth, bloating;

From the cardiovascular system: tachycardia, increased blood pressure, pain in the heart, arrhythmias;
From the respiratory system: very rarely – shortness of breath, suffocation attacks.
Allergic reactions: hypersensitivity reactions, fever, very rarely – angioedema.
From the skin: pruritus, hyperemia of the skin, urticaria.
Interaction
Thiotriazoline as a cardioprotective drug can be used in combination with basic agents for the treatment of coronary heart disease.
As a hepatoprotective agent, it can be combined with the appointment of traditional methods of treating hepatitis of the appropriate etiology.
How to take, course of use and dosage
Parenterally.
In chronic hepatitis, Thiotriazoline is administered intramuscularly in the first five days of the disease,2 ml of a 2.5% solution 2-3 times a day (2-3 times 50 mg), or intravenously slowly, at a rate of 2 ml / min,4 ml of a 2.5% solution (100 mg), or dropwise at a rate of 20-30 drops per minute (2 ampoules of a 2.5% solution are dissolved in 150-250 ml of saline).
From the fifth to the twentieth day of treatment, Thiotriazoline is prescribed in tablets (100 mg 3 times a day).
The course of treatment is 20-30 days.
